Cloning and vector production for transfection of prokaryotic and eukaryotic cells

Cloning into transfection plasmids, with or without reporter genes is offered starting from cDNAs and plasmids provided by the customer or by accessing cDNAs and plasmids provided by COMT. This service includes the production of mutated constructs by site-directed mutagenesis of amino acid residues, constructs carrying specific deletions or constructs containing fragments of the cDNA of the gene of interest.

"Gene profiling" analyses
-
Conventional DNA microarray-based gene expression profiling on costumer-supplied cellular or tissue samples is offered relying upon the Agilent platform. One-color (a sample per array) and two-color (two samples per array) experiments are optionally offered. Statistics is based upon 2 spots/gene run in duplicate or triplicate.
The standard protocol entails:
- extraction and purification of total RNA;
- qualitative and quantitative analysis of RNA extracted by microfluidic systems;
- fluorescent labelling of RNA with specific fluorophores and quantification of labelled RNA;
- hybridization and slides image acquisition;
- data processing.

Functional gene analysis by RNA interference (RNAi)
-
Protein knockdown by RNA interference technology, using naked or vector-contained siRNA or shRNA probes selected by COMT or based upon sequences provided by the customer. The service includes the design and validation of at least one functional probe and of specific control-probes directed to other segments of the RNA target. Upon request, it is also possible to receive “phenotype rescuing analyses” by BACs transfection or transfection of a specific recombinant plasmid.
